Need advice, inherited cat that is not adjusting well.

Hello r/Pets. I have a difficult situation. A close friend of my family passed and left 2 cats behind. I took these cats in because I didn't like the alternative of putting them in a shelter or them being separated. They are sisters and get along well but one of the cats (Calico) is not adjusting. We've had them both for 6 months. The Black cat is progressing as well as one would expect. We can't pick her up but she does explore the house periodically and will sit next to me/husband, unprompted (as in giving her treats). She is still shy/timid but over the course of months doesn't always run to her hiding spot. The Calico constantly hides, actively runs away from us, pees outside the litter box and cries a lot. We have tried Feliway plug ins, calming collars, sprays. Nothing seems to alleviate her anxiety/fear. We've taken her to the vet (a month ago) and were instructed to give her medicine but it isn't helping (she refused to eat, no matter how fine we smash it into her food).

We sit quietly, we give her treats sometimes, to encourage her to come to us. She comes but is always jumpy. They share a room that we set up for them, it's a 12x12ft space, with a patio area, where they can perch and look out, over our back yard. The Calico will usually jump over/run past us to get to other parts of the house only to cry and mark anything she can sniff. They each have a set of toys, but only the Black cat will play with them. I really don't know what else to do, is this a sign that she does not like us as owners? Any assistance, on this matter, would be much appreciated.
